Injured in the hand, the Czech's last tournament was at Wimbledon, where she was title-holder and lost in the first round.
Down to 39th in the world, she had to use a protected ranking to enter the Adelaide draw.
On Monday, she came up against Anastasia Pavlyuchenkova and won 6-4, 6-7, 6-2.
Of particular note was her remarkable serving performance, with 22 aces. Vondrousova will also be at the Australian Open the following week.
